Investigational fixed-dose combination of the amylin analog cagrilintide and the GLP-1 agonist semaglutide from Novo Nordisk, developed to deliver additive weight loss beyond either alone.
01 Overview
CagriSema is a once-weekly combination of cagrilintide, a long-acting amylin receptor agonist, and semaglutide, a GLP-1 receptor agonist. The rationale is that amylin and GLP-1 act through complementary satiety pathways, producing greater weight loss together.
Phase 3 REDEFINE trials reported substantial weight loss, though somewhat below early expectations and with variability depending on dose adherence. Because it combines an approved GLP-1 agent with an investigational amylin analog, its overall evidence base is mid-stage and still developing.
02 Mechanism
Combines amylin receptor agonism (cagrilintide) with GLP-1 receptor agonism (semaglutide) for additive appetite suppression and slowed gastric emptying.
