Tesofensine
An investigational triple monoamine reuptake inhibitor (serotonin, noradrenaline, dopamine) originally studied for Parkinson's and Alzheimer's disease, then repurposed for obesity after weight loss was noticed in trials. Phase II data showed strong weight loss, but it is not approved anywhere and remains investigational with dose-dependent cardiovascular and mood concerns.
01 Overview
Tesofensine (NS2330) inhibits reuptake of serotonin, noradrenaline and dopamine, an unusually broad monoaminergic profile that suppresses appetite. Its earlier neurodegenerative-disease trials failed, but a Phase II obesity trial (TIPO-1) reported placebo-subtracted weight loss of around 9-10% at the top dose over 24 weeks — larger than approved oral drugs of its era.
Despite that efficacy it has not reached the market; higher doses raised heart rate and blood pressure and produced mood/psychiatric and sleep effects, and later development stalled. It appears in the grey/research-chemical market, where quality and safety are unverified.
