Pioglitazone
Pioglitazone (Actos) is a thiazolidinedione PPAR-gamma agonist that improves insulin sensitivity by promoting fat storage in subcutaneous adipose and improving glucose uptake. Bodybuilders occasionally use it as a partitioning agent, but water retention, weight gain and bone/heart-failure concerns limit its appeal.
01 Overview
Pioglitazone activates PPAR-gamma in adipose tissue, driving adipocyte differentiation and redistributing lipid from visceral and ectopic depots into subcutaneous fat, which improves whole-body insulin sensitivity. It is an approved type 2 diabetes drug with substantial trial data.
Some physique users adopt it hoping to improve nutrient partitioning, but its pronounced fluid retention and weight gain are counterproductive for a lean look, and it carries warnings for heart failure, fractures and a debated bladder-cancer signal.

Agonises the nuclear receptor PPAR-gamma, upregulating genes for adipocyte differentiation, lipid uptake and glucose transport, thereby improving peripheral insulin sensitivity and lowering circulating free fatty acids.
