Oxytocin
A nine-amino-acid neurohypophyseal hormone that drives uterine contraction and milk let-down, and modulates social and affiliative behaviour. Long-established as an injectable obstetric drug (labour induction, postpartum haemorrhage); intranasal oxytocin is widely studied for social cognition but remains investigational there.
01 Overview
Oxytocin is synthesised in the hypothalamus and released from the posterior pituitary. Its peripheral roles — stimulating uterine smooth muscle during labour and the myoepithelial cells of the breast for milk ejection — are the basis of its decades-old licensed use as an intravenous or intramuscular agent in obstetrics.
A large research literature examines intranasal oxytocin for effects on trust, bonding, anxiety and autism-spectrum social deficits. Results are mixed and often fail to replicate, and no central-nervous-system indication is approved. The obstetric evidence is strong; the neuropsychiatric/behavioural evidence is genuinely uncertain, so effects there are rated accordingly.
02 Mechanism
Agonist at the oxytocin receptor (a Gq-coupled GPCR) on uterine and mammary smooth muscle and in CNS circuits governing social and stress behaviour.
