Landogrozumab (LY2495655)
Landogrozumab (LY2495655) is a humanised anti-myostatin monoclonal antibody studied by Eli Lilly for sarcopenia, post-surgical muscle loss and cancer cachexia. Trials showed increases in lean mass and some functional measures in older adults, but overall benefits were modest and development did not continue to approval.
01 Overview
Landogrozumab neutralises myostatin to counter age- and illness-related muscle loss. In older adults after hip-fracture surgery and in other populations, treated groups gained appendicular lean mass and showed improvements in some physical performance tests.
The magnitude of functional benefit was limited and inconsistent across endpoints, and the antibody was not advanced to registration. Its data set is one of the more informative for myostatin blockade in ageing rather than genetic muscle disease.
02 Mechanism
A humanised IgG4 monoclonal antibody against mature myostatin, blocking its interaction with activin type II receptors and thereby increasing muscle mass.
