Hexarelin
Hexarelin is a synthetic hexapeptide growth-hormone-releasing peptide (GHS-R agonist), more potent than GHRP-6 at releasing GH. It has been studied in small human trials for GH secretion and cardiovascular effects, but tachyphylaxis (desensitisation with continued use) and cortisol/prolactin stimulation limit it. Non-medical physique use is anecdotal.
01 Overview
Hexarelin strongly stimulates GH release and has independent cardioprotective actions at the CD36 and GHS receptors seen in preclinical and small clinical studies. It raises cortisol and prolactin somewhat, and GH response attenuates with sustained daily use as the receptor desensitises.
Because of desensitisation and the cortisol/prolactin effects it is used less than ipamorelin for ongoing body-composition goals. Human data exists for acute GH-releasing potency but not for physique endpoints.
02 Mechanism
Potent agonist of the growth-hormone secretagogue receptor (GHS-R1a) and CD36, evoking a strong GH pulse; also modestly raises ACTH/cortisol and prolactin, with receptor desensitisation on repeated dosing.
