FOXO4-DRI
An experimental senolytic peptide designed to disrupt the interaction between FOXO4 and p53, selectively pushing senescent ('zombie') cells into apoptosis. Known from a single influential mouse study; there is no approved human use and no controlled human trial data.
01 Overview
FOXO4-DRI (D-Retro-Inverso) is a cell-penetrating peptide engineered to competitively block FOXO4 binding to p53 inside senescent cells. In aged and damaged mice, periodic dosing cleared senescent cells and improved markers of fitness, renal function and fur density, generating substantial interest in longevity research.
Despite widespread grey-market sale, FOXO4-DRI has essentially no published human safety or efficacy data. All effect claims derive from rodent and in-vitro experiments, and its selectivity, dosing and long-term consequences in humans are unknown.
02 Mechanism
A D-retro-inverso peptide that disrupts the FOXO4-p53 interaction in senescent cells, releasing p53 to trigger targeted apoptosis (senolysis).
