Bupropion
An atypical antidepressant and smoking-cessation aid (Wellbutrin, Zyban) that inhibits dopamine and noradrenaline reuptake. Combined with naltrexone in Contrave for weight management. Low abuse potential and no weight gain, but lowers seizure threshold and can cause insomnia and cardiovascular effects.
01 Overview
Bupropion is a noradrenaline-dopamine reuptake inhibitor used for major depression and smoking cessation, and as the stimulant component of the weight-loss combination Contrave (bupropion/naltrexone). Unlike many antidepressants it tends to be weight-neutral or cause modest weight loss and does not cause sexual dysfunction.
It is not a controlled substance and has low abuse potential, though misuse has been reported rarely. Its most important risk is a dose-dependent lowering of the seizure threshold, contraindicating it in seizure disorders, bulimia/anorexia and abrupt alcohol withdrawal. Insomnia, dry mouth, agitation and modest blood-pressure increases are common.
02 Mechanism
Inhibits reuptake of dopamine and noradrenaline and antagonises nicotinic acetylcholine receptors, without significant serotonergic activity.
