alpha-PVP (flakka)
alpha-PVP (alpha-pyrrolidinopentiophenone, 'flakka') is a pyrrolidine synthetic cathinone and potent dopamine/noradrenaline reuptake inhibitor. It gained notoriety from clusters of severe agitation, hyperthermia and excited-delirium presentations. Human knowledge is from emergency toxicology, case series and animal pharmacology rather than trials; it is markedly more compulsive than the ring-substituted cathinones.
01 Overview
alpha-PVP is one of the pyrrolidine cathinones, a subgroup that acts as strong catecholamine reuptake inhibitors rather than releasers. Effects are intensely stimulating with little of the warmth of mephedrone-type compounds, and the very high potency and short duration make it especially prone to compulsive, binge redosing. It is used orally, insufflated, and (outside this profile's scope) smoked.
Media attention followed reports of agitated, psychotic and hyperthermic presentations. Documented harms include extreme cardiovascular strain, severe agitation and psychosis, hyperthermia, and a severe comedown. Long-term neurotoxicity is unconfirmed. As a pyrrolidine cathinone it is flagged as especially compulsive.
02 Mechanism
Potent and relatively selective dopamine and noradrenaline reuptake inhibitor (DAT/NET), producing strong stimulation with high abuse liability and low serotonergic activity.
03 Dosing
| Tier | Dose | Route | Notes |
|---|---|---|---|
| Light | 3–10 mg | Insufflated | Rapid, intense onset; strong compulsion to redose. |
| Light | 5–15 mg | Oral | Very potent; active in single-milligram amounts. High overdose risk. |
| Common | 15–30 mg | Oral | Narrow margin; compulsive redosing common. |
