4-Methylmethamphetamine (4-MMA)
4-Methylmethamphetamine (4-MMA) is a ring-methylated methamphetamine analogue with substantial serotonin-releasing activity. Its pharmacology resembles the para-methylated 'toxic' amphetamines (PMA/PMMA family in effect profile), and it has been associated with serotonergic toxicity and deaths. Human data is very limited.
01 Overview
4-MMA is methamphetamine with a methyl group added to the aromatic ring at the para position. This substitution markedly increases serotonergic activity, giving it a profile more hazardous than methamphetamine and reminiscent of para-substituted amphetamines known for serotonin toxicity.
There is little controlled human data; knowledge derives from animal studies, analytical case reports and monitoring of deaths involving related compounds. The combination of stimulant and strong serotonergic action raises the risk of hyperthermia and serotonin toxicity.
02 Mechanism
Releases dopamine, noradrenaline and, prominently, serotonin. The para-methyl group shifts activity toward serotonin release, associated with hyperthermia and serotonin toxicity.
